WebMechanistically, metformin induces energy stress through inhibition of complex I of the respiratory chain in mitochondria. This leads to a change in the ATP-to-AMP ratio and canonical AMPK activation. There has been extensive research to evaluate the contribution of AMPK to the effect of metformin on circulating glucose and lipids.
